In humans, however, allantoin forms only via non‑enzymatic oxidation triggered by free radicals. Studies in APP/tau mutant mice show that urinary allantoin levels rise before Alzheimer’s disease symptoms appear, but not in early or middle disease stages, suggesting its potential as a predictive biomarker for disease onset. Because allantoin in humans is formed under free‑radical stress, elevated urinary concentrations serve as a useful biomarker for systemic oxidative stress status.
