Acetaminophen is a widely used analgesic and antipyretic agent. Unlike most non‑steroidal anti‑inflammatory drugs that block both COX‑1 and COX‑2, earlier research found it has relatively weak inhibitory activity against both isoforms. However, ex vivo tests in human blood show it inhibits COX‑2 by 83% and COX‑1 by 56% at a 1000 mg dose, with IC50 values of 25.8 μM and 113.7 μM respectively. Acetaminophen is converted both enzymatically and non‑enzymatically into several reactive metabolites that may cause side effects, including liver damage.

Acetaminophen, internationally known as Paracetamol, is a common analgesic and antipyretic drug without anti‑inflammatory or anti‑rheumatic effects. Its fever‑reducing action is comparable to aspirin, while its analgesic effect is moderate. It is a leading aniline derivative and is especially suitable for people who cannot tolerate carboxylic acid drugs. It is used to relieve cold symptoms and toothache. It also serves as an intermediate in organic synthesis, a stabilizer for hydrogen peroxide, and a photographic chemical.
