Schisandrol B is a type of lignan compound first extracted from Schisandra chinensis, which possesses significant hepatoprotective properties. It upregulates the expression of pregnane X receptor (PXR) target genes related to bile acid metabolism. In mouse liver, these genes include Cyp3a11, Ugt1a1, Oatp2, and Mrp3; in HEK293T cells, they cover CYP3A4, UGT1A1, and OATP2. It effectively prevents hepatic necrosis and intrahepatic cholestasis caused by lithocholic acid in wild-type mice, but not in Pxr-knockout mice. At a dosage of 100 mg/kg twice daily, it reduces mortality in cholestasis mouse models. It also accelerates liver regeneration after partial hepatectomy and mitigates hepatotoxicity caused by acetaminophen.
